Category:

Finished Products

HA35 Topical Comfort Gel

Description

Net Weight: 25 g

Key Ingredient: 10% high-concentration bioactive hyaluronan formulation

Developed for localized topical care of areas affected by superficial pain, burning, stinging, redness, swelling, and inflammatory discomfort.

Directions for Use

Apply an appropriate amount of gel evenly to the affected area. For prolonged contact, the treated area may be covered with an appropriate occlusive film to minimize drying.

Published Observations

In a published 4-patient case series evaluating high-concentration HA35 gel for herpes zoster–related pain:

● Mean pain scores decreased by approximately 55% within 2 minutes

● Pain scores decreased further to approximately 1–2 within 20 minutes

● Reported pain relief generally persisted for approximately 4 hours

● In one patient with postherpetic neuralgia, occlusive application extended reported relief to more than 8 hours

● Redness and other local inflammatory signs also improved during follow-up

Mechanistic Research

Experimental electrophysiology studies showed that HA35 modulates the sensory ion channels TRPV1 and TRPA1, which are involved in pain, itch, and inflammatory signaling.

This provides a potential mechanistic basis for the rapid sensory responses observed following topical HA35 application.

Oral Absorption & Distribution

HA35 & HA70 · Preclinical oral absorption study

Oral administration studies in rats compared two defined low-molecular-weight HA fractions, HA35 and HA70.

Key findings:

• Both HA35 and HA70 were rapidly detected in systemic circulation

• HA levels in the mesenteric lymph nodes increased significantly within 30 minutes after administration

• Mesenteric lymphatic transport was identified as a major absorption pathway

• Free HA showed no comparable increase in the liver or spleen during the observation period

These findings support direct intestinal uptake of defined low-molecular-weight HA fragments and highlight the mesenteric lymphatic system as an important route connecting oral administration with systemic exposure.

Separate radiotracer imaging studies have also detected orally administered 99mTc-labeled HA35 in mesenteric and other lymph nodes following gastrointestinal administration.

• Mean observed gingival coverage increase: approximately 1.2 mm

• Chronic gingival / radicular pain score: 2.4 → 0

Additional observations included:

• Gingival itching and discomfort disappeared within approximately 30 seconds after application

• Redness and swelling improved markedly within 4 days

• Brushing-associated bleeding stopped within 2 days

• No adverse reactions were reported

These observations were reported in a small uncontrolled case series.

D. Mechanistic Support — TRPV1 & TRPA1

Patch-clamp electrophysiology · n=12 per condition

At 400 μg/mL:

• HA35 inhibited capsaicin-activated TRPV1 currents by 34.46% (P < 0.0001)

• HA35 inhibited AITC-activated TRPA1 currents by 29.27% (P < 0.001)

• No significant effect was observed on TRPV3

These findings provide a molecular link between HA35 and sensory ion-channel pathways involved in pain and itch.
