35kDa玻尿酸片段的止痛、止痒和抑制组织炎症的作用机制
35kDa 玻尿酸片段 HA35 主要通过淋巴循环途径而非血液循环,加速局部炎症的消退与吸收

基于人体感觉系统的敏感性,海诺实验室首席科学家惠觅宙教授和美国罗切斯特大学的Gorbunova教授,发现10%高浓度35kDa玻尿酸片段10秒消除皮肤瘙痒和30秒内消除1厘米左右深的表浅疼痛(1、2、3、4、5)。惠觅宙与 Gorbunova 教授提出了一项创新性猜想:这些快速起效的治疗效果,并非依赖 35kDa 玻尿酸片段分子直接渗入人体组织而产生 —— 这一猜想为传统药物动力学与药效学理论补充了革命性的全新视角。
瞬间受体电位TRP通道广泛分布于皮肤或黏膜表面的神经末梢,其核心功能是秒级快速介导温度、痛觉和痒觉信号传递。例如,TRPV1和TRPA1是感知热/痛/痒的瞬间受体电位TRP(6、7)。高浓度35kDa玻尿酸片段HA35通过与皮肤或黏膜表面接触,以非分子渗透的方式直接抑制表皮表面的瞬间受体电位TRP,阻止疼痛和瘙痒信号经神经末梢传导进入人体组织深处, 产生秒级止痛和止痒作用。这一过程中,瞬间受体电位TRP细胞膜电位,在10-30秒内实现痒觉和痛觉信号的阻断。
瞬间受体电位TRPV1和TRPA1还存在于角质形成细胞、免疫细胞等非神经细胞中,高浓度35kDa玻尿酸片段HA35通过调控这些细胞的TRPV1和TRPA1,还能抑制炎症,从源头减少炎症性疼痛。
此外,我们的研究还证实,35kDa 玻尿酸片段 HA35 经皮下注射后,可通过与淋巴内皮细胞表面的玻尿酸 LYVE-1 受体特异性结合,在 1-5 分钟内快速进入体表淋巴结,并融入淋巴循环系统(8、9、10)。不仅如此,HA35 还能促进新鲜提取的单个核细胞(以淋巴细胞为主)向模拟炎症区域的胶滴外扩散,有效减少炎症细胞在局部炎症组织的聚集与浸润(8、9)。综上所述,35kDa 玻尿酸片段 HA35 主要通过淋巴循环途径而非血液循环,加速局部炎症的消退与吸收。
