Description
HA70 is a low-molecular-weight bioactive hyaluronan fraction with an average molecular weight of approximately 70 kDa, with more than 80% of its molecular-weight distribution within the 30–90 kDa range.
Biological Activity
● A 10% high-concentration HA70 formulation rapidly relieves skin itching within approximately 10 seconds.
● A 10% high-concentration HA70 formulation rapidly relieves superficial pain at a depth of approximately 1–2 cm within 30 seconds.
● HA70 passes through a 220 nm pore-size filtration membrane, demonstrating size-dependent permeability.
● At concentrations of 3 mg/mL or lower, HA70 interacts with the surface of red blood cells and induces characteristic coin-like erythrocyte aggregation.
● At 300 μg/mL, HA70 promotes the outward migration and diffusion of inflammatory cells, predominantly lymphocytes, from gel droplets simulating inflamed tissue.
● At 400 μg/mL, HA70 inhibits the sensory ion channels TRPV1 and TRPA1, which are associated with pain, itch, and inflammatory signaling.
● Following subcutaneous administration in mice, radiolabeled HA70 rapidly enters nearby lymph nodes and the spleen, as demonstrated by molecular imaging.

Oral Absorption & Distribution
HA35 & HA70 · Preclinical oral absorption study
Oral administration studies in rats compared two defined low-molecular-weight HA fractions, HA35 and HA70.
Key findings:
• Both HA35 and HA70 were rapidly detected in systemic circulation
• HA levels in the mesenteric lymph nodes increased significantly within 30 minutes after administration
• Mesenteric lymphatic transport was identified as a major absorption pathway
• Free HA showed no comparable increase in the liver or spleen during the observation period
These findings support direct intestinal uptake of defined low-molecular-weight HA fragments and highlight the mesenteric lymphatic system as an important route connecting oral administration with systemic exposure.
Separate radiotracer imaging studies have also detected orally administered 99mTc-labeled HA35 in mesenteric and other lymph nodes following gastrointestinal administration.
